NSC624206 is an inhibitor of ubiquitin E1 (UBA1), with an IC50 of ~9 μM. NSC624206 specifically blocks ubiquitin-thioester formation (IC50=13 μM) but has no effect on ubiquitin adenylation.
体外研究：
NSC624206 prevents in vitro ubiquitination of the cell cycle regulator p27 as well as inducing an accumulation of p27 in the HepG2 liver cancer cell line.NSC624206 clearly reduces ubiquitin-E1 thioesters in a concentration-dependent manner with an IC50 of 13 μM.NSC624206 (10 nM-10 μM; 24 hours) induces accumulation of p27 in cells.
