MRK-740 is a potent, selective and substrate-competitive PRDM9 histone methyltransferase inhibitor with an IC50 of 80?nM. MRK-740 is more selective for PRDM9 than other histone methyltransferases and other non-epigenetic targets. MRK-740 reduces PRDM9-dependent trimethylation of H3K4 (IC50?=?0.8?μM).
体外研究：
处理 24 小时后，MRK-740 (3 μM) 在 IC90 时不影响 HEK293T 细胞生长，但在 10 μM 时观察到一些毒性。MRK-740 是 MCF7 细胞中 H3K4 甲基化的等效抑制剂。在细胞中，MRK-740 特异性地直接抑制内源性 PRDM9 靶位点的 H3K4 甲基化。
